A case of a differential lysis for female infidelity A husband suspected his wife's infidelity and delivered a pair or her panties. Upon completion of the positive semen detection (PSA81996) and Y-PLEX DNA analysis (FR2008864) the wife claimed that the panties were their daughter's. The customer then ordered a differential lyses and the PowerPlex profiles clearly demonstrated that one male and one female was present in the sample. The customer then delivered a floss pick of his wife's. The floss pick matched the female profile from the panties. Subsequently a Private Investigator employed by the customer delivered a water bottle from the suspected adulterer. The bottle had to be processed under a different serial number due to the legal chain of custody being intact. The water bottle profile from the suspect was identical to the male profile obtained from the differential lyses. Case # FR2008864 (GQ0018836) Evidence The items to be tested are R1-swab packet labeled “Customer” containing two swabs and R2-one 1.7 mL tube labeled “sperm sample” cloth punches. Analysis DNA extraction, PCR and Genotyper was performed on sample R1 and sample R2. In addition, a Y-PLEX PCR analyzing the alleles DYS389I, DYS439, DYS438, DYS437, DYS392, DYS393, DYS19, DYS389II, DYS390, DYS391, and DYS385 was also performed on the samples R1 and R2. Results:
Conclusions: Based
on the analysis of the alleles DYS389I, DYS439, DYS438, DYS437,
DYS392, DYS393, DYS19, DYS389II, DYS390, DYS391, and DYS385, the
profiles obtained from Samples R1 and R2 are consistent with
male DNA. The profiles for Sample R1 and Sample R2 are not
identical. Analysis Report Case # FR2008866 (GQ0018848 / GQ0018851) Evidence The items to be tested is Sample R1 FR2008866R1- 1.7 ml with a small piece of cloth labeled “Differentail Lysis from Panties; Sample R2 FR2008866R2- labeled “Floss pick”. Analysis A differential lysis was performed on Sample R1 FR2008866R1 to separate sperm and epithelial cells in different groups allowing a profile for any contributors to Sample R1 FR2008866R1 to be profiled. DNA extraction, PCR and Genotyper was preformed on Sample R1 FR2008866R1 and Sample R2 FR2008866R2. Results:
Conclusions: Based on the analysis of the alleles D3S1358, THO1, D21S11, D18S51, Penta E, D5S818, D13S317, D7S820, D16S539, CSF1PO, Penta D, vWA, D8S1179, TPOX, and FGA, the above profile were obtained for Sample R1F FR2008866r1f, Sample R1M FR2008866r1m and Sample R2 FR2008866R2. Sample R1 FR2008866r1 female enriched cell fraction is identical to Sample R2 FR2008866R2. Sample R1 FR2008866R1 and R2 FR2008866R2 were consumed in testing.Analysis Report Case # FR2008869 (GQCA18891) vidence The item to be tested is R1- one envelope containing a 1.7 mL tube containing a swab of a water bottle. Analysis DNA extraction, PCR and Genotyper were performed on the Sample R1.
Conclusions: Based on the analysis of the alleles CSF1PO, D2S1338, Penta E, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, Penta D, D21S11, FGA, THO1, TPOX, and vWA, a single male DNA profile was obtained for Sample R1.
